The Duplica Prep is an automated nucleic acids purification system that allows to obtain DNA and RNA from different specimens types. Duplica Prep. is a IVD CE “walk away” equipment that provides pure nucleic acids with a high yield of recovery suitable for diagnostic and research purposes. Duplica Prep. platform avoids cross contamination problems, by the use of disposable tips, which prevent the contact with biological samples.

Duplica Prep Overview

Principle of functioning

The principle of functioning of the Duplica Prep platform is based on the nucleic acids separation through the use of magnetic beads. The system has been developed during several years through the collaboration with clinical diagnostic laboratories.

The platform operates in association with the panel of reagents Duplica Extraction Kit, this synergy allows to obtain, with good yield, nucleic acids (DNA and RNA) suitable for molecular biology applications. The platform includes a barcode reader useful for a better samples traceability.

The processing time is variable between 40 - 75 minutes depending on the extraction matrices. The number of samples processed varies from 1 to 12 without reagents wasting.

The magnetic separation is made through the use of 12 metal rods that are magnetized by an external magnet. The magnetization is required to capture the beads and to transfer them into the subsequent solutions. During the steps in which the rods are not magnetized, the rods play an efficient role of beads resuspension into the extraction solution (washing and elution solutions). The resuspension is made by the rods rotation at high speed. This fast and efficient phase, plays a major role in obtaining good yield and quality of nucleic acids (DNA and RNA) (see picture 1).

It’s finally relevant to underline that, the use of Duplica Prep’s technology allows to avoid liquid transfer, since the system captures and moves the beads from one buffer to another. This feature, together with the use of disposable tips, allows to virtually prevent any contamination problem.

Generic Features for the Duplica Prep Automated DNA/RNA Extraction System:

No Cross Contamination
Duplica Prep extraction platform presents no cross contamination problems, because the equipment components are never in contact with the biological samples. The magnetic rods are protected through the use of a single-use special tips, patented for this platform.

Walk Away System
Each extraction set up requires an hands-on part, concerning the positioning of the 96 Deep Well Plate (supplied together with the extraction reagents) and Tip and Tube Rack for tips loading, on the internal truck. After the extraction start, the operator intervention is no required until the end of the extraction procedure.

Lack of Inhibition Problems
The reagents associate to the Platform include 4 different washing buffers. Several washing steps are performed in order to guarantee the elimination of inhibition problems.

Extraction Control
During the extraction set up, the system allows the inclusion of an extraction control, if required by the operator.
